Effects of Squalene on the Skin of Aged Mice: Light Microscopic and Laser Confocal Microscopic Study

Abstract
To evaluate the effects of squalene (SQ) on skin of aged mice, SQ was applied to the back skin of a 10 month-old ICR female mouse at 12 hour-intervals for 10 days. The expression of epidermal growth factor (EGF) and histologic alterations on the SQ-treated skin using under the of a light microscope and laser confocal microscopes were performed. In the light microscopic observation, the development of a germ cell layer was weak in the normal aged skin, the number of the cells, however, started to increase after 2 days. The increase in the number of epidermal cells was directly proportional to the period of SQ treatment and the whole cell layers in epidermis were easily differentiated after 5 days of SQ treatment In the laser confocal microscopic observation, the activity of EGF was increased with time after 24 hours of SQ treatment. In summary, the application of SQ to the skin of aged mice could induce the increase in skin tissue expression of EGF, and promote germ cell differentiation. Therefore, it is suggested that SQ plays a major role in differentiation or renewal of skin tissue in aged mice.
